Long-term effects of moderate elevation of oxidation–reduction potential on European seabass (Dicentrarchus labrax) in recirculating aquaculture systems ArchiMer
Li, Xian; Przybyla, Cyrille; Triplet, Sebastien; Liu, Ying; Blancheton, Jean-paul.
The long term effects of moderate elevation ORP (oxidation–reduction potential) around 300–320 mV on the growth, hematological parameters and the ability of European seabass (Dicentrarchus labrax) to react against bacterial infection was studied in recirculating aquaculture systems (RASs). Two RASs, one with a moderate ozonation (RAS-O3) and a control (RAS-C) were used in this experiment. After 60 days, seabass reared in the RAS-O3 were more able to react against a Vibrio anguillarum infection. It was in spite of the fact that seabass in the RAS-O3 showed decreased feed intake, feed conversion rate, growth rate and modified hematological parameters compared with the fish in RAS-C. It is obvious that an ORP level of 300–320 mV is too high for seabass to...

Measurement of the isotopic composition of dissolved iron in the open ocean ArchiMer
Lacan, F.; Radic, A.; Jeandel, C.; Poitrasson, F.; Sarthou, Geraldine; Pradoux, C.; Freydier, R..
This work demonstrates for the first time the feasibility of the measurement of the isotopic composition of dissolved iron in seawater for a typical open ocean Fe concentration range (0.1-1 nM). It also presents the first data of this kind. Iron is preconcentrated using a Nitriloacetic Acid Superflow resin and purified using an AG1x4 anion exchange resin. The isotopic ratios are measured with a MC-ICPMS Neptune, coupled with a desolvator (Aridus II), using a Fe-57-Fe-58 double spike mass bias correction. Measurement precision (0.13%, 2SD) allows resolving small iron isotopic composition variations within the water column, in the Atlantic sector of the Southern Ocean (from delta Fe-57 = -0.19 to +0.32 parts per thousand). Isotopically light iron found in...

Mesure de la demande chimique en oxygène dans l'eau de mer. ArchiMer
Michel, Pierre.
Introduction: Measuring the chemical oxygen demand (COD) is a way of determining the organic matter in water based on its oxidisibility by bichromate. The presence of organic matter in sea water is one of the factors that determines the productivity of the oceans: whether this organic matter will be absorbed directly or must first be transformed into nutritive substances by bacteria. This beneficial action also has its limits. On the one hand, consumption of organic matter remains limited; when it exceeds a certain rate, there is no longer any increase in productivity. On the other hand, the excess organic matter deteriorates by consuming the dissolved oxygen and thus competes with marine life. In extreme cases, this excess can cause the eutrophication of...

Multi-Element Determination of Trace Elements in Natural Water Reference Materials by ICP-SFMS after Tm Addition and Iron Co-precipitation ArchiMer
Bayon, Germain; Birot, Dominique; Bollinger, Claire; Barrat, Jean Alix.
We report on an improved method for determining trace element abundances in seawater and other natural waters. The analytical procedure involves co-precipitation on iron hydroxides after addition of a Tm spike, and measurement by inductively coupled plasma-sector field mass spectrometry (ICP-SFMS). The validity of the method was assessed through a series of co-precipitation experiments, using ultra-diluted solutions of a certified rock reference material (BIR-1). Results obtained for four natural water reference materials (NASS-5, CASS-4, SLEW-3, SLRS-4) are in agreement with published working values for rare earth elements, yttrium, vanadium and, when available, for hafnium, zirconium, thorium and scandium. A set of proposed values with uncertainties...

Natural Degradation and Biodegradation of Poly(3-Hydroxybutyrate-co-3-Hydroxyvalerate) in Liquid and Solid Marine Environments ArchiMer
Deroine, Morgan; Cesar, Guy; Le Duigou, Antoine; Davies, Peter; Bruzaud, Stephane.
In this study, natural degradation and biodegradation of poly(3-hydroxybuyrate-co-3-hydroxyvalerate) (PHBV) films were followed in different marine environments. First of all, ageing of PHBV films was investigated in natural seawater for 180 days and degradation was followed by means of weight loss measurements, scanning electron microscopy (SEM), differential scanning calorimetry and steric exclusion chromatography. In a second part, biodegradation tests were performed on PHBV powder, by following carbon dioxide (CO2) release(,) to highlight the PHBV bioassimilation of marine microorganisms. Three different marine environments were considered for biodegradation tests: a solid inoculum with foreshore sand, a solid-liquid inoculum with sand and seawater and...

Riquelme,Javiera; Olaeta,José Antonio; Gálvez,Lena; Undurraga,Pedro; Fuentealba,Claudia; Osses,Andrea; Orellana,Jaime; Gallardo,José; Pedreschi,Romina.
Sarcocornia neei is a halophyte that grows on the coast of the Valparaiso Region of Chile. Studies related to its nutritional and functional value under wild and cultivated conditions are not available. Thus, in this study, a nutritional (complete proximal, mineral and dietary fiber analysis) and functional characterization (total phenolics, β-carotene, hydrophilic and lipophilic antioxidant activity (HAA and LAA) and ascorbic acid) were performed for wild and cultivated plants. Wild plants displayed higher amounts of compounds involved in stress defense mechanisms such as total phenolics, proteins, dietary fiber and ash. The mineral analysis revealed that Na+ and Cl- are the main ions accumulated in wild and cultivated Sarcocornia neei that are...

Pasteurization: A reliable method for preservation of nutrient in seawater samples for inter-laboratory and field applications ArchiMer
Daniel, Anne; Kerouel, Roger; Aminot, Alain.
Following previous work, the production of reference material for nutrients in seawater, using pasteurization as a preservation method, was carried out seven times between 2006 and 2010 in the framework of inter-laboratory exercises. The preparation of samples from natural seawater allowed to become depleted in nutrients then spiked, bottled and pasteurized, is described. Five main nutrients are involved in this study: ammonium, nitrite, nitrate, phosphate and silicate. Bottles are in glass for the ammonium samples and in plastic for the other nutrients. Pasteurization was performed at 80 ± 3 °C for 2 h. Samples were controlled for homogeneity and stability at 3–5 month interval, before and after the period allocated for each exercise and a third time...

Performances of dead-end ultrafiltration of seawater: from the filtration and backwash efficiencies to the membrane fouling mechanisms ArchiMer
Masse, Anthony; Arab, Oumnia; Sechet, Veronique; Jaouen, Pascal; Pontie, Maxime; Sabiri, Nour-eddine; Plantier, Severine.
The present work investigates the fouling mechanisms of PVDF hollow fibre membrane (0.03 μm) during the dead end ultrafiltration at a fixed permeate flux (outside to inside configuration) of complex synthetic seawater composed by humic acids, alginic acids, inorganic particles and numerous salts at high concentrations. Short term ultrafiltration experiments at 100 L.h-1.m-2 show that the optimal specific filtered volume seems to be equal to 50 L.m-2. A residual fouling resistance equal to 2.1010 m-1 is added after each cycle of filtration during 8h of ultrafiltration at 100 L.h-1.m-2 and 50 L.m-2. Most of the fouling is reversible (80%). Organics are barely (15% of humic acids) retained by the membrane. Backwash efficiency drops during operation which...

Polychloroprene behaviour in a marine environment: Role of silica fillers ArchiMer
Tchalla, Siwavi Tatiana; Le Gac, Pierre-yves; Maurin, R.; Creac'Hcadec, R..
Polychloroprene rubbers are widely used in marine structures and often filled with silica in order to increase mechanical properties. The presence of silica fillers leads to a complex degradation of the material. This study aims to understand the deeper degradation mechanisms involved when a silica filled polychloroprene is used in sea water. To do so, 4 polychloroprene rubbers filled with different amounts of silica (from 0 to 45 phr) were aged in natural sea water for 6 months at temperatures ranging from 25 to 60 °C. Moreover, a natural rubber with similar formulation was also considered in order to evaluate the role of the chlorine atom in the degradation. . The chemistry and mechanics of the rubber degradation were also studied. In the presence of...

Réaction de réduction de l'oxygène sur les aciers inoxydables en eau de mer naturelle. Influence du biofilm sur les processus de corrosion ArchiMer
Le Bozec, Nathalie.
iofilm development on stainless steels immersed in natural seawater can have prejudicial consequences on the resistance of these materials to corrosion . The goal of the present study was to get more precise information on the corrosion processes, and especially on the oxygen reduction reaction. As the reaction is linked to the stainless steel surface state, the characterisation of the oxides films (composition, structure, thickness...) is essential to understand the mechanisms and the oxygen reduction kinetic. The first aim of the study has been to correlate the oxygen reduction processes with the characteristics of the oxides layer as a function of the alloy surface treatment (mechanical polishing, electrochemical passivation and prereduction, chemical...

Responses of enteric bacteria to environmental stresses in seawater ArchiMer
Troussellier, Marc; Bonnefont, Jean-luc; Courties, Claude; Derrien, Annick; Dupray, Elizabeth; Gauthier, Michel; Gourmelon, Michele; Joux, Fabien; Lebaron, Philippe; Martin, Yvan; Pommepuy, Monique.
The effects of different environmental factors (nutrient deprivation, hyperosmotic shock, exposure to light) on enteric bacteria which have been transferred into the marine environment, have been studied experimentally (microcosms) by considering demographic, physiological and genetic responses in Escherichia coli or Salmonella typhimurium populations. Short-term experiments (less than or equal to 48 h) showed that nutrient deprivation induced limited changes in measured bacteriological variables, but when combined with hyperosmotic shock, it results in an energy charge decrease and inactivation of membrane transport. Light exposure mainly affects the colony-forming capacity of bacterial populations. Combining different stress factors confirmed the rapid...

Rotavirus VLP2/6: a new tool for tracking rotavirus in the marine environment ArchiMer
Loisy, Fabienne; Atmar, Robert L.; Cohen, Jean; Bosch, Albert; Le Guyader, Soizick.
The potential of rotavirus 2/6-virus-like-particles (VLP2/6) for use as tracers in the marine environment was investigated. The stability of bovine rotavirus (strain RF) and VLP2/6 in natural seawater at 25degreesC for six days was studied. ELISA and western blot methods were used to quantify the particles. The rates of decline of rotavirus particles and VLP2/6 were similar (approximately 0.5 log 10 per day). Western blot analysis showed that the integrity of capsid proteins VP2 and VP6 was conserved during the incubation time. These results demonstrate that VLP2/6 particles have the same stability in seawater as rotavirus particles. Thus, VLP2/6 can be used as a tracer, which should be of particular value for studying the fate of rotavirus particles in...

ABSTRACT This review focuses on the potential of halophytes for food, fodder and biofuels production, as well as their impacts on the environment and societies. Moreover, to open new areas in production systems using novel technologies such as halophytes in a desert agriculture. We are faced with the crisis and the shortage of freshwater in arid, semi-arid and desert regions. For this reason, we have to apply sustainable systems for human food, fodder and biofuels. Halophytes are naturally resistant to salt and develop on the coastal coast and arid-saline areas. We present a complete summary of the current situation of human population growth and food demand, a sustainable alternative such as halophilic crops of agro-industrial importance compared with...

Sanitary quality of sands from marine recreational beaches of São Paulo, Brazil BJM
Sato,Maria Inês Zanoli; Di Bari,Marisa; Lamparelli,Claudia Condé; Truzzi,Ana Cristina; Coelho,Maria Cristina L.S.; Hachich,Elayse Maria.
A sanitary evaluation of sand and water from 16 beaches of São Paulo State, Brazil, was undertaken during spring of 1997 and summer of 1998. Ninety six samples each of wet and dry sand and seawater were collected and analysed for fecal indicator bacteria. A parasitological examination and Candida albicans analysis were also performed in sand samples and F-specific bacteriophages were determined in seawater. Statistical analysis of the results demonstrated higher concentrations of fecal coliforms and fecal streptococci in dry sand during summer. Correlation analysis indicated a significant relationship between fecal indicator densities in wet sand and seawater. There was a significant correlation between the densities of fecal coliforms and fecal...

Seawater accelerated ageing of poly(3-hydroxybutyrate-co-3-hydroxyvalerate ArchiMer
Deroine, Morgan; Le Duigou, Antoine; Corre, Yves-marie; Le Gac, Pierre-yves; Davies, Peter; Cesar, Guy; Bruzaud, Stephane.
The aim of this study is to establish a baseline for poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) lifetime prediction in a marine environment, by means of mechanical and physico-chemical characterization of specimens immersed in continuously renewed and filtered natural seawater in the laboratory at different temperatures (4, 25 and 40°C). Samples were also aged at sea in Lorient harbour in order to compare laboratory and natural degradation mechanisms and kinetics. Due to its morphology, hydrolysis of PHBV in natural seawater is quite slow, and samples were observed to undergo preferentially an enzymatic surface degradation. Increasing the aging temperature in the laboratory promotes the water uptake and causes hydrolysis. As two degradation...

Sulfide determination in hydrothermal seawater samples using a vibrating gold micro-wire electrode in conjunction with stripping chronopotentiometry ArchiMer
Aumond, Virginie; Waeles, Mathieu; Salaun, Pascal; Gibbon-walsh, Kristoff; Van Den Berg, Constant M. G.; Sarradin, Pierre-marie; Riso, Ricardo D..
A rapid electrochemical stripping chronopotentiometric procedure to determined sulfide in unaltered hydrothermal seawater samples is presented. Sulfide is deposited at -0.25 V (vs Ag/AgCl, KCl 3 M) at a vibrating gold microwire and then stripped through the application of a reductive constant current (typically -2 mu A). The hydrodynamic conditions are modulated by vibration allowing a short deposition step, which is shown here to be necessary to minimize H2S volatilization. The limit of detection (LOD) is 30 nM after a deposition step of 7s. This LOD is in the same range as the most sensitive cathodic voltammetric technique using a mercury drop electrode and is well below those reported previously for other electrodes capable of being implemented in situ....

The determination of total dissolved free primary amines in seawater: Critical factors, optimized procedure and artefact correction ArchiMer
Aminot, Alain; Kerouel, Roger.
The paper reviews the methods described in the literature for the determination of total dissolved free primary amines (TDFPA) by fluorescence. A wide set of reaction conditions can be found, but they rely on few experiments for their validation. Among fluorogenic compounds, o-plithaldialdehyde (OPA) is more sensitive than fluorescamine and was thus examined here. However, the use of mercaptoethanol (ME) in the reaction (as ail additional derivatization compound) is able to generate unreliable results, in particular when standardization relies on glycine. We suggest replacing ME with 3-mercaptopropionic acid (MPA) which induces more stable and comparable fluorescence among amine Compounds. A systematic study was therefore undertaken to define reagent...
